The factors influencing the selection of a subset of infected individuals to be reactivated and progress to clinical disease are not well understood, and as a result, no FDA approved vaccine exists for the prevention of tuberculosis disease. Understanding how M. tuberculosis establishes latency and reactivation from latency is crucial to discovering strategies to prevent tuberculosis. This information will be gained through analysis of the relationships between M. tuberculosis infection and host genetics, bacterial gene expression and the host immune response. We hypothesize that specific bacterial gene expression signatures associated with bacterial growth phase, growth rate, and host genetics will provide an interpretable and specific signature for each phase of M. tuberculosis infection, allowing us to predict the progression of infection to disease. Specifically, bacteria that are non-replicating or contained in small granulomas will express different types of stress response and growth phase genes, and expression of particular combinations of genes will be indicative of the phase of infection in which bacteria are. We will characterize the relationships between host genetics, bacterial gene expression, and host immune response during each phase of M. tuberculosis infection using the mouse as a model of infection. In order to define and validate bacterial gene expression signatures associated with each phase of M. tuberculosis infection, we will perform a series of experiments in the mouse.
